Knockdown of TLR9 promotes cell autophagy to ameliorate LPS-induced inflammation of HK-2 cells. Notes: After sepsis cell models were transfected with si-TLR9, qRT-PCR and western blot were employed to examine the transfection efficiency of si-TLR9 (A, B). Additionally, the HK-2 cells were subjected to 3-MA for 30 min, exposed to 1 µg/mL LPS and then transfected with si-TLR9. The immunofluorescence was utilized for measurement of fluorescence intensity of LC3 (C), TEM for observation of the number of autophagosomes (D), and western blot (E) for analysis of protein expression levels of autophagy-related proteins LC3-II/LC3-I (F), p62 (G), and Beclin-1 (H).
